Researchers from New York University have employed advanced techniques, including holographic microscopy and super-resolution imaging, to explore biomolecular condensates. These are tiny, membraneless compartments within cells that play a crucial role in cellular functions. This research marks the first time scientists have been able to measure the protein content and growth dynamics of individual biomolecular condensates without disturbing them.
Saumya Saurabh, assistant professor of chemical biology at NYU and senior author of the study published in the Journal of the American Chemical Society, said: “It’s been the elephant in the room for scientists. Our research provides a precise and non-invasive way to study biomolecular condensates.”
David Grier, professor of physics and director of the Center for Soft Matter Research at NYU, commented on their findings: “Being able to see ‘under the hood’ for the first time has revealed some big surprises about this important class of systems.”
Biomolecular condensates concentrate specific molecules like proteins and nucleic acids without being enclosed by a membrane. The process known as phase separation is essential for organizing cellular biochemistry. Although traditionally studied using fluorescent tags or two-dimensional surfaces, these methods often disturb their behavior due to their sensitivity.
Julian von Hofe, a PhD candidate in Saurabh’s group and first author on the study, noted: “I was surprised by their complex and incredibly sensitive response to different ionic species. Even a small change in ionic valency drastically altered both condensate concentration and dynamics.”
The team overcame traditional challenges by developing a system that allows real-time examination through holographic microscopy. Grier’s lab has pioneered this technique using lasers and lenses to create three-dimensional images captured on video.
The researchers applied this label-free method to study PopZ protein-based condensates critical for cell growth. They measured protein concentration inside these structures by determining single protein volume inspired by Benjamin Franklin’s oil slick experiment.
Super-resolution imaging further revealed that these condensates exhibit intricate nanoscale organization rather than being simple uniform droplets.
Grier stated: “Our collaboration has introduced fast, precise, and effective methods for measuring the composition and dynamics of macromolecular condensates.”
Understanding biomolecular condensate organization could provide insights into diseases such as ALS or neurological disorders where proteins transition from fluid states into harmful plaques.
Saurabh explained: “In a disease like ALS… understanding how a spherical condensate forms into a deadly plaque is an opportunity to better understand ALS.” Furthermore, drug molecules often end up inside these structures affecting efficacy due to sequestration within them potentially causing side effects when targeting specific proteins.
“With this new approach… we can now explore chemical space… achieving specificity needed preventing drugs entering unwanted areas,” said Saurabh regarding future drug design possibilities avoiding unintended interactions within cellular environments potentially reducing adverse reactions associated with treatment options available today across various medical conditions requiring therapeutic intervention based upon scientific advancements made possible through continued exploration innovative methodologies developed collaboratively between experts fields chemistry physics biology
